[目的]分析硫酸化相关酶在AIS患者关节突软骨中的表达特点,为软骨细胞的力学研究和延缓脊柱侧凸进展提供参考。[方法]选取18例Lenke 1型AIS患者顶椎和下端椎凹、凸侧和对照组(15例)的下关节突软骨作为研究对象,固定脱钙后制成石蜡切片。免疫组化染色测定分析硫酸化反应相关酶PAPSS2、PAPST1和CHST1在Lenke 1型AIS患者顶椎和下端椎关节突软骨中的表达,利用Leica显微镜对免疫组化图像进行观察,用Image-pro Plus软件将每个视野中的着色面积、光密度以及目标数目转换成累积光密度值(IOD),代表相应酶的表达强度,最后进行统计学分析。[结果]PAPSS2主要分布在软骨细胞质和软骨囊基质;PAPST1特异性分布在软骨细胞质;CHST15主要分布在软骨细胞质和软骨囊基质。硫酸化反应相关酶PAPSS2、PAPST1和CHST15的表达转换成IOD有以下分布特点:对照组两侧的表达差异无统计学意义;顶椎凹侧高于凸侧,顶椎凸侧高于对照组,差异有统计学意义;顶椎高于同侧下端椎,差异有统计学意义;下端椎的凹凸侧的表达差异无统计学意义;下端椎的表达与对照组的表达比较差异无统计学意义。[结论]青少年特发性脊柱侧凸顶椎关节突硫酸化反应相关酶的表达增强与特发性脊柱侧凸进展相关。
[Objective] To analyze the expression characteristics of sulfated enzymes in articular cartilage of patients with AIS, and provide reference for the mechanics of chondrocytes and delay the progression of scoliosis. [Methods] 18 cases of Lenke type 1 AIS patients with vertebral and inferior vertebrae, the convex side and the control group (15 cases) of the lower articular cartilage were selected as the research object, the fixed decalcification made of paraffin section. Immunohistochemical staining was used to analyze the expression of PAPSS2, PAPST1 and CHST1 in the vertebral and inferior vertebral articular cartilage of Lenke type 1 AIS patients. Immunohistochemistry was performed using a Leica microscope. Image-pro Plus The software converts the colored area, optical density, and target number in each field of view to the cumulative optical density value (IOD), which represents the intensity of expression of the corresponding enzyme, and finally performs a statistical analysis. [Results] PAPSS2 mainly distributed in cartilage cytoplasm and cartilaginous matrix; PAPST1 specific distribution in cartilage cytoplasm; CHST15 mainly distributed in cartilage cytoplasm and cartilage matrix. The expression of sulfatase-related enzymes PAPSS2, PAPST1 and CHST15 converted into IOD has the following distribution characteristics: the expression of the two sides of the control group showed no significant difference; the concave side of the apical vertebra was higher than the convex side, the convex vertebra was higher than the control group, The difference was statistically significant; the vertebral vertebra was higher than the ipsilateral lower vertebra, the difference was statistically significant; the expression of inferior vertebra was not statistically significant; the expression of lower vertebra was not significantly different from the control group. [Conclusion] The increased expression of enzymes related to sulfonation of idiopathic scoliosis in adolescents is associated with the progression of idiopathic scoliosis.
