目的:实验通过大黄虫丸含药血清干预体外人肺腺癌PAa细胞株研究其抗肿瘤机制,进而探讨活血化瘀法在肿瘤治疗中的疗效和价值。方法:采用血清药理学方法以不同浓度的大黄虫丸含药血清与人肺腺癌PAa细胞株共同孵育,于不同时间观察,并与空白血清作对照。研究大黄虫丸含药血清对体外人肺腺癌PAa细胞株的细胞毒作用、生长抑制作用及对细胞周期的影响。结果:5%~20%各组含药血清OD值与相同作用时间10%牛血清、相同浓度的正常鼠血清OD值比较有明显差异(P<0.05~0.01),并随着血药浓度的提高OD值逐渐减低;药血清对人肺腺癌PAa细胞株增殖有明显抑制作用,并随药血清浓度提高、作用时间延长抑制作用增强;其诱导凋亡作用随药血清浓度的增加及作用时间的延长而增强。结论:大黄虫丸含药血清对体外人肺腺癌PAa细胞株有明显抑制作用,为大黄虫丸抗肿瘤提供了科学依据。
OBJECTIVE: To investigate the antitumor mechanism of Rhubarb worm pill-containing serum in human lung adenocarcinoma PAa cell line in vitro and to explore the therapeutic effect and value of activating blood circulation and removing blood stasis method in tumor therapy. Methods: Serum pharmacology method was used to co-culture PAH cell lines with different concentration of Dahuang Zhechong pill and observed with different time, and compared with blank serum. To investigate the cytotoxicity, growth inhibition and cell cycle of Rhubarb and Chronic Obstructing Medicine-containing serum on human lung adenocarcinoma PAa cell line in vitro. Results: The serum OD value of 5% ~ 20% of each group was significantly different from that of 10% bovine serum of the same duration and the same concentration of normal mice (P <0.05 ~ 0.01). With the increase of plasma concentration Increase OD values gradually reduced; serum on human lung adenocarcinoma PAa cell proliferation was significantly inhibited, and with the serum concentration increased, the role of prolonged inhibition increased; the role of apoptosis induced by serum concentration and duration of action The extension of the increase. Conclusion: Rhubarb Zhechong pill containing serum on human lung adenocarcinoma PAa cell lines in vitro significantly inhibited, providing a scientific basis for anti-tumor Dahuang Zhechong pill.
