目的 探讨Clara细胞在慢性阻塞性肺疾病 (COPD)发病中的作用。方法 收集COPD患者肺手术标本 13例 ,并收集年龄、吸烟指数 (吸烟支数 /天×年 )相近的非COPD患者 13例肺标本 ,免疫组化法观察小气道中Clara细胞所占比例及分布 ,RNA酶保护实验测定肺组织中CC16mRNA表达水平 ,Western法测定CC16蛋白水平。结果 COPD组呼吸性细支气管中Clara细胞数目低于非COPD组 (P <0 0 5 ) ,肺组织中CC 16mRNA的表达低于非COPD组 (P <0 0 5 ) ,CC16蛋白水平在两组中差异无显著性。结论 Clara细胞的数目与功能的变化可能是COPD形成的机制之一
Objective To investigate the role of Clara cells in the pathogenesis of chronic obstructive pulmonary disease (COPD). Methods Thirteen lung specimens from COPD patients were collected and 13 lung specimens from non-COPD patients with similar age and smoking index (smoking count / day × year) were collected. The proportion and distribution of Clara cells in the small airway were observed by immunohistochemistry, The level of CC16 mRNA in lung tissue was determined by RNase protection assay, and the level of CC16 protein was determined by Western blot. Results The number of Clara cells in the bronchioles of COPD group was lower than that in non-COPD group (P <0.05). The expression of CC 16 mRNA in lung tissue was lower than that in non-COPD group (P <0.05) In the difference was not significant. Conclusion The change of number and function of Clara cells may be one of the mechanisms of COPD formation
