目的环氧化酶(COX)-2抑制剂(塞来西布)对大鼠烫伤创面组织中COX-2和VEGF的表达的影响。方法将90只SD大鼠随机分为A(10只)、B(40只)、C(40只)组。A组为空白对照组,B、C组予以氯胺酮麻醉,10%硫化钠脱毛,将大鼠背部浸入90℃水中10s,造成面积为20%的深Ⅱ度烫伤模型。三组均给予正常饲料喂养,C组伤后给予塞来西布0.4mg/(g.d),共7d。B、C组于伤后1、3、5、7、14d取创面组织标本,A组在同样时间点取背部组织标本。用免疫组化SABC法观察标本中环氧化酶-2(COX-2)和血管内皮生长因子(VEGF)的表达情况。结果A组各时点COX-2、VEGF表达相同。伤后1、3、5d,B组COX-2及VEGF的表达逐渐增高,C组COX-2及VEGF均有所下降(P<0.05);伤后7、14d,B组COX-2表达下降而VEGF表达增加,C组与B组相比COX-2、VEGF的表达较低。结论COX-2抑制剂可抑制创面组织中COX-2、VEGF的表达,COX-2和VEGF在创面的愈合中发挥着重要作用。
Effect of cyclooxygenase - 2 inhibitor (Celecoxib) on the expression of COX - 2 and VEGF in burned wound of rats. Methods Ninety SD rats were randomly divided into A (n = 10), B (n = 40) and C (n = 40) groups. Group A was blank control group. Rats in groups B and C were anesthetized with ketamine and ephedrine with 10% sodium sulfide. The rats were dipped in water at 90 ℃ for 10 s, resulting in a deep second degree scald model with an area of 20%. All the three groups were fed with normal feed, Group C was given Celecoxib 0.4mg / (g.d) for 7 days. Group B and C received wound tissue samples at 1, 3, 5, 7 and 14 days after injury, and group A received back tissue samples at the same time points. The expression of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) were observed by immunohistochemical SABC method. Results The expression of COX-2 and VEGF were the same in group A at each time point. At 1, 3 and 5 days after injury, the expression of COX-2 and VEGF in group B gradually increased, while the expression of COX-2 and VEGF decreased in group C (P <0.05) While the expression of VEGF increased. The expression of COX-2 and VEGF in C group was lower than that in B group. Conclusions COX-2 inhibitor can inhibit the expression of COX-2 and VEGF in wound tissues. COX-2 and VEGF play an important role in wound healing.
