目的观察长期注射大剂量同种异体抗角蛋白自身抗体(AK auto Ab)后,家兔血清中针对抗角蛋白自身抗体(AK auto Ab)独特型抗体的产生。方法肌肉注射亲和层析纯化的AK auto Ab(5 mg/kg),隔日1次,连续90 d,用ELISA检测血清中的抗独特型抗体。结果注射AK auto Ab的家兔产生了针对AK auto Ab F(ab′)2片段的抗独特型抗体,于给药4 wk血清抗体滴度达高峰,以后逐渐降低。结论长期注射大剂量同种异体AK auto Ab的家兔可产生免疫耐受性。
Objective To observe the production of idiotypic anti-keratin antibodies (AK auto Ab) in rabbits serum after long-term injection of high-dose alloantibody anti-keratin autoantibodies (AK auto Ab). Methods AK auto Ab (5 mg / kg) purified by affinity chromatography was injected intramuscularly once a day for 90 consecutive days. Serum anti-idiotypic antibodies were detected by ELISA. Results Rabbits injected with AK auto Ab produced anti-idiotypic antibodies against the AK auto Ab F (ab ’) 2 fragment, which peaked at 4 weeks after administration of the antibody and then gradually decreased. Conclusion Long-term injection of large doses of allogeneic AK auto Ab in rabbits produces immunological tolerance.
