赵忠新黄坚庄建华邵福源实验科张玲珍报道选择31例符合Poser诊断标准的多发性硬化(MS)患者及20例正常人为对照组为研究对象.MS复发期23例,男10例,女13例,年龄16～53岁,平均(34.1±9.2)岁;缓解期8例,男3例,女5例,年18～41岁,平均(30.9±7.7)岁.MS和对照组除外标准:其他自身免疫性疾病、肿瘤、近期感染、组织损伤、3个月内应用免疫抑制剂.用流式细胞仪检测血和脑脊液(CSF)Fas和FasL的表达,Fas用直接法;全血或CSF 100μL,分别加入FITC标记的Fas单抗,孵育30min;FasL用间接法:全血或CSF 100μL分别加入FasL单抗,孵育30min后,PE标记的二抗孵育30min,经溶红细胞、稀释和固定后,24 h内上流式细胞仪分析.操作中根据不同白细胞前向和侧向光散射的不同可区分淋巴、单核和中性粒细胞.每个样本分析5000个细胞,结果以阳性细胞的百分率表示.rn
