本实验用离体豚鼠右心房标本,以心率为指标观察了青藤碱对Iso和His量效反应及CaCl_2时效反应的影响。青藤碱可使离体右房的Iso和His量效曲线非平行右移,并伴随最大反应的降低,呈非竞争性拮抗作用,pD′_2值分别为3.48±0.08、3.63±0.04,分别不同于β-受体阻断剂心得安及H_2-受体阻断剂甲氰咪胍。同时,青藤碱对CaCl_2所致的正性频率有明显的拮抗作用,与戊脉安相似。提示青藤碱的负性心率作用不是通过阻断β-受体和H_2-受体,而可能是阻滞Ca~(2+)通道的结果。
In this study, isolated guinea pig right atrium specimens to heart rate as an indicator of sinomenine on the Iso and His quantitative response and CaCl 2 aging response. Sinomenine induced a non-parallel shift of the Iso and His dose-response curves of right atrial isolated rightward with a decrease of maximal response, showing a noncompetitive antagonistic effect with pD’_2 values of 3.48 ± 0.08 and 3.63 ± 0.04 respectively Different from β-blockers propranolol and H_2-receptor blocker cimetidine. At the same time, sinomenine had a significant antagonistic effect on the positive frequency induced by CaCl 2, which was similar to that of verapamil. It is suggested that the negative effect of sinomenine on heart rate may not be the result of blocking the Ca 2+ channel by blocking the β-receptor and H 2 -receptor.
