目的检测十堰地区不同Hoehn-Yahr分级的散发性帕金森病患者血浆a-共核蛋白水平的差异。方法选择2012年9月-2015年1月在湖北医药学院附属太和医院就诊的48例原发性帕金森病患者,参照Hoehn-Yahr分级标准分组,对照组选择年龄、性别均与病例组匹配的健康人。采用ELISA方法检测比较48例帕金森病患者与48例正常对照组以及不同Hoehn-Yahr分级PD患者血浆中a-synuclein蛋白寡聚体水平的差异。结果十堰地区48例散发性帕金森病患者血浆中a-synuclein蛋白单体水平与对照组相比无显著差异,PD组寡聚体水平(3.869±1.292 ng/m L)与对照组(2.922±0.804 ng/m L)比较有显著差异,且不同Hoehn-Yahr分级的PD患者血浆a-synuclein蛋白寡聚体水平比较有显著性差异(P<0.05)。结论在十堰市我们首次开展了散发性帕金森病患者血浆a-synuclein寡聚体水平高于正常人,且随着疾病严重及分期的进展,其水平呈上升趋势。因此,血浆a-synuclein蛋白寡聚体可作为帕金森病诊断的有潜力的生物标志物,为协助帕金森病诊断及疾病分期提供帮助。
Objective To detect the plasma a-synuclein levels in patients with sporadic Parkinson’s disease with different Hoehn-Yahr grading in Shiyan. Methods Seventy-eight patients with primary Parkinson’s disease who were treated in Taihe Hospital Affiliated to Hubei Medical College from September 2012 to January 2015 were selected according to the Hoehn-Yahr classification criteria. The age and sex of the control group were matched with the case group Healthy people. The difference of α-synuclein protein oligomer in 48 patients with Parkinson’s disease and 48 normal controls and different Hoehn-Yahr grade PD patients was detected by ELISA. Results There was no significant difference in plasma a-synuclein protein levels in 48 patients with sporadic Parkinson’s disease in Shiyan. The level of oligomer in PD group (3.869 ± 1.292 ng / m L) was significantly higher than that in control group (2.922 ± 0.804 ng / m L), and there was a significant difference in plasma a-synuclein protein oligomers between PD patients with different Hoehn-Yahr grading (P <0.05). Conclusions We first conducted a population of patients with sporadic Parkinson’s disease in Shiyan City, plasma a-synuclein oligomer levels higher than normal, and with the severity of disease and staging, the level is on the rise. Therefore, the plasma a-synuclein protein oligomer can be used as a potential biomarker for the diagnosis of Parkinson’s disease, to assist Parkinson’s disease diagnosis and disease staging.
