内脂素对人胆管癌细胞QBC939侵袭及增殖影响的体外研究

目的:研究人内脂素(visfatin)对胆管癌细胞QBC939体外增殖和侵袭能力的影响,并初步探讨可能存在的机制。方法:培养胆管癌细胞QBC939,在不同浓度的内脂素作用不同时间后,以四甲基偶氮唑盐(MTT)法检测细胞增殖能力,用Tran-swell小室检测内脂素(200 ng/ml)24 h后对胆管癌QBC939细胞侵袭能力的影响,Western blot检测内脂素对QBC939细胞内基质金属蛋白酶(matrix metalloproteinase,MMP)-2蛋白的表达。结果:内脂素可以促进QBC939细胞的增殖(P<0.01),且存在剂量依赖性;内脂素(200 ng/ml)处理24 h后,QBC939细胞穿透聚碳酸酯膜的能力与对照组相比显著增强(P<0.01);Westernblot显示内脂素(200 ng/m1)处理24 h后,QBC939细胞MMP-2蛋白的表达明显增高。结论:内脂素可以明显提高人胆管癌细胞QBC939的增殖和侵袭能力,其作用机制可能与MMP-2水平的增高有关。 Objective: To study the effect of visfatin on the proliferation and invasion of cholangiocarcinoma QBC939 in vitro and to explore the possible mechanism. Methods: The cultured cholangiocarcinoma cells QBC939 were cultured in different concentrations of visfatin for different time. Cell proliferation was measured by MTT assay. The visfatin (200 ng / ml) for 24 h on the invasion ability of QBC939 cells. Western blot was used to detect the expression of matrix metalloproteinase (MMP) -2 protein in QBC939 cells. Results: Florescein promoted the proliferation of QBC939 cells (P <0.01) in a dose-dependent manner. After treated with 200 ng / ml visfatin for 24 h, the ability of QBC939 cells to penetrate through the polycarbonate membrane was significantly lower than that of the control group (P <0.01). Western blot showed that the expression of MMP-2 protein in QBC939 cells was significantly increased after treated with 200 ng / ml visfatin for 24 h. Conclusion: Florescein can significantly improve the proliferation and invasion of human cholangiocarcinoma QBC939 cells, and its mechanism may be related to the increase of MMP-2 level.
