目的观察围产期母鼠在长期高蛋氨酸饮食的情况下,对子代大鼠离乳后的免疫功能的影响。方法 Wistar孕鼠,6只,以添加1.8%蛋氨酸的标准AIN-93饲料喂食,直至产后第25天,建立高蛋氨酸饮食组;6只Wistar孕鼠喂以普通饲料作为正常对照。计数大鼠分娩时仔鼠数量、体重;子代大鼠出生第25天离乳,次日,取子代大鼠静脉血、胸腺、脾脏,记录胸腺指数、脾指数,流氏细胞仪计数血CD4+、CD8+T细胞,Elisa法检测血浆肿瘤坏死因子(TNF-α)、白细胞介素(IL-6)水平,HE染色观察脾脏大体病理改变,实时荧光定量PCR法检测脾脏TNF-α、IL-6、NF-κB基因表达的改变。结果妊娠期、哺乳期母鼠在长期进食高蛋氨酸饮食后,孕鼠产仔数量减少,仔鼠体重偏低,子代大鼠外周血CD4+T细胞计数(P<0.05)、CD8+T细胞计数、CD4+/CD8+比值(P<0.05)、胸腺指数(P<0.05)、脾指数(P<0.05)明显下降,血浆TNF-α、IL-6升高(P<0.05),脾脏发育不良,脾TNF-α、IL-6、NF-κB mRNA表达增加(P<0.05)。结论围产期母鼠在长期进食高蛋氨酸饮食后,产仔数量减少,子代大鼠免疫功能显著低于正常大鼠。
Objective To observe the influence of perinatal rats on the immune function after off-lactation of offspring in the condition of long-term high methionine diet. Methods Six Wistar pregnant rats were fed with standard AIN-93 diet supplemented with 1.8% methionine until the 25th day after giving birth to a high-methionine diet. Six Wistar pregnant rats were fed normal diet. The offspring of the offspring were born on the 25th day after birth, and the venous blood, thymus and spleen of offspring rats were taken the next day, and the thymus index, spleen index and flow cytometry count blood The levels of tumor necrosis factor (TNF-α) and interleukin (IL-6) were measured by Elisa method. Results During pregnancy and lactation, the number of littermates, the weight of offspring mice, the CD4 + T cell count (P <0.05) and the number of CD8 + T cells (P <0.05), the ratio of CD4 + / CD8 + (P <0.05), thymus index (P <0.05) and spleen index The expressions of TNF-α, IL-6 and NF-κB mRNA in spleen increased (P <0.05). Conclusions The number of littermates in perinatal dams after long-term high-methionine diet decrease and the immune function in offspring rats is significantly lower than that in normal rats.
