目的:研究瑞舒伐他汀钙对高尿酸血症大鼠血清尿酸、C反应蛋白、白细胞介素-6、肿瘤坏死因子-α和主动脉内皮功能的影响。方法:60只大鼠随机分为5组,每组各12只,分别为对照组,模型组,瑞舒伐他汀钙高、中、低剂量组。模型组和瑞舒伐他汀高、中、低剂量组采用氧嗪酸钾联合酵母膏建立高尿酸血症模型,瑞舒伐他汀钙高、中、低剂量组在建立模型同时分别给予瑞舒伐他汀钙10.0,5.0,2.5 mg/(kg.d)治疗6周,观察各组大鼠血清尿酸、C反应蛋白、白细胞介素-6及肿瘤坏死因子-α的水平和主动脉内皮病理形态学变化。结果:瑞舒伐他汀钙高、中、低剂量组血清C反应蛋白、白细胞介素-6、肿瘤坏死因子-α及血尿酸水平均明显低于模型组(P<0.01)。病理结果显示主动脉内皮损害模型组较对照组严重,瑞舒伐他汀钙高、中、低剂量组血管内皮损害呈剂量依赖性减轻,以高剂量组损害最轻。血清C反应蛋白、白细胞介素-6、肿瘤坏死因子-α水平变化与血清尿酸水平呈正相关。结论:高尿酸血症大鼠存在炎症反应,可损伤主动脉内皮细胞功能;瑞舒伐他汀钙有抑制炎症反应,保护主动脉内皮细胞功能的作用,可降低血尿酸,呈剂量依赖性。
Objective: To investigate the effect of rosuvastatin calcium on serum uric acid, C-reactive protein, interleukin-6, tumor necrosis factor-α and aortic endothelial function in hyperuricemic rats. Methods: Sixty rats were randomly divided into five groups, each containing 12 rats, which were control group, model group and rosuvastatin calcium high, medium and low dose groups respectively. In model group and rosuvastatin high, medium and low dose groups, potassium oxonate and yeast extract were used to establish hyperuricemia model. The pathological results showed that the model group of aortic endothelial injury was more serious than the control group, and the damage of vascular endothelial cells in high, medium and low dose rosuvastatin calcium group was relieved in a dose-dependent manner, and the lightest in high dose group. Serum C-reactive protein, interleukin-6, tumor necrosis factor-α levels and serum uric acid levels were positively correlated. CONCLUSION: Hyperuricemia rats have inflammatory reaction, which can damage the function of aortic endothelial cells. Rosuvastatin calcium can inhibit the inflammatory reaction and protect the function of aortic endothelial cells, and can reduce the blood uric acid in a dose-dependent manner.
