本文采用体细胞杂交瘤技术和酶联免疫吸附试验筛选技术,获得了6株持续分泌抗人甲状腺球蛋白 McAb 的杂交瘤细胞系。这6种 McAb 对人 T_3/T_4、及家兔、小牛、大鼠的甲状腺球蛋白均无交叉反应,但其中4种 McAb 对狗甲状腺球蛋白有不同程度的交叉反应,包括2种 McAb对人甲状腺微粒体也有交叉反应,余2种 McAb 均无交叉反应。将所得杂交瘤细胞接种 BALB/c鼠腹腔,获得高效价的腹水 McAb。本文还以这些 McAb 用免疫组化法,研究和检测病理甲状腺组织中甲状腺球蛋白的可能性进行了初步探索,并讨论了其临床意义。
In this study, 6 hybridoma cell lines secreting anti-human thyroglobulin McAb were obtained by using somatic hybridoma technique and enzyme-linked immunosorbent assay. There was no cross-reaction between the six McAbs against human T_3 / T_4 and thyroglobulin in rabbits, calves and rats, but the four McAbs had different degrees of cross-reactivity with dog thyroglobulin, including two McAbs Human thyroid microsomes also have cross-reaction, no cross-reaction of the other two McAbs. The obtained hybridoma cells were inoculated into the abdominal cavity of BALB / c mice to obtain high titer McAbs of ascites. In this paper, we also used these McAbs immunohistochemical method to study and detect the possibility of thyroglobulin in pathological thyroid tissue was initially explored and discussed its clinical significance.
