目的观察白藜芦醇对家兔心房肌、乳头肌的影响。方法测定白藜芦醇对家兔离体右心房窦性频率的影响,并研究白藜芦醇在不同类型钾通道阻断剂存在情况下对心脏负性变频作用的影响;测定白藜芦醇对家兔离体乳头肌收缩力的影响。结果白藜芦醇可剂量依赖性地减慢右心房窦性频率(P<0.05),1×10-3mol/L白藜芦醇可使右心房窦性频率减慢(9.4±3.5)%;与白藜芦醇组相比,ATP敏感性钾通道(KATP)阻断剂格列苯脲(glibenclam ide,G li)可部分阻断白藜芦醇的作用,其负性变频作用由原来的(9.4±3.5)%降为(7.1±1.0)%(P<0.05),但钙激活性钾通道(KCa)阻断剂四乙胺(tetraethylammonium,TEA)、电压依赖性钾通道(KV)阻断剂4-氨基吡啶(4-am inopyrid ine,4-AP)、内向整流性钾通道(KIR)阻断剂氯化钡(barium chloride,BaC l2)、乙酰胆碱(Ach)调节钾通道(KAch)阻断剂阿托品(atropine)不能阻断白藜芦醇的负性变频作用(P>0.05)。白藜芦醇对乳头肌收缩力的影响不显著(P>0.05)。结论白藜芦醇可呈剂量依赖性减慢右心房窦性频率,其作用与开放KATP通道有关,与KCa、KV、KIR和KACh通道无关。白藜芦醇对心室乳头肌无明显的负性肌力作用。
Objective To observe the effect of resveratrol on atrial and papillary muscles in rabbits. Methods The effect of resveratrol on the frequency of isolated right atrial sinus in rabbits was determined. The effect of resveratrol on the negative frequency conversion of cardiac in the presence of different types of potassium channel blockers was investigated. Resveratrol was determined. Effects on the contractility of isolated papillary muscles in rabbits. Results Resveratrol slowed the right atrial sinus frequency in a dose-dependent manner (P<0.05), and 1×10-3 mol/L resveratrol reduced the right atrial sinus frequency by (9.4±3.5)%. Compared with the resveratrol group, ATP-sensitive potassium channel (KATP) blocker glibenclamide (G li) partially blocks the effect of resveratrol, and its negative frequency conversion effect is (9.4 ± 3.5)% decreased to (7.1 ± 1.0)% (P <0.05), but the calcium-activated potassium channel (KCa) blockers tetraethylammonium (TEA), voltage-dependent potassium channel (KV) resistance Breakers 4-aminopyridine (4-AP), inward rectifier potassium channel (KIR) inhibitors barium chloride (BaCl 2), acetylcholine (Ach)-regulated potassium channels (KAch) The blocker atropine did not block the negative conversion of resveratrol (P > 0.05). Resveratrol had no significant effect on the contractility of papillary muscles (P>0.05). Conclusion Resveratrol can reduce the frequency of right atrial sinus in a dose-dependent manner. The effect of resveratrol is related to the opening of KATP channels and has nothing to do with KCa, KV, KIR and KACh channels. Resveratrol has no significant negative inotropic effects on the ventricular papillary muscles.
