利用匀强电场对杉木当年生茎段外植体进行处理,研究电场强度和处理时间对杉木茎段芽诱导的影响。在1.0 k V·cm-1的电场强度下处理5 min,芽诱导率最高,达100%,比对照组增加62.50%;芽形成能力达835.00%,是对照组的2.78倍,亦高于其它处理组合;平均芽数最高达到12.50个,是对照组的2.50倍,与对照组具有显著差异。7.0 k V·cm-1电场强度处理1 min后,随着处理时间的增加,诱导率均为0,明显抑制了杉木芽的分化;处理3 min后,外植体存活率均呈下降趋势,致死作用增大。结果表明:电场强度和处理时间对杉木外植体的芽诱导率和形成能力具显著影响。
The effect of electric field intensity and treatment time on the induction of stem segments of Chinese fir was studied by using the uniform electric field to treat the stem explants of Chinese fir. Under the electric field of 1.0 kV · cm-1 for 5 min, the bud induction rate was the highest, reaching 100%, 62.50% higher than that of the control group. The bud formation ability reached 835.00%, which was 2.78 times higher than that of the control group Treatment combination; the average number of shoots up to 12.50, 2.50 times that of the control group, with the significant difference between the control group. After treated with 7.0 k V · cm-1 electric field strength for 1 min, the induction rate was 0 with the increase of treatment time, significantly inhibited the differentiation of Chinese fir shoots. After 3 min of treatment, the survival rate of explants showed a downward trend, Lethal effect increases. The results showed that the electric field intensity and the treatment time significantly affected the shoot induction rate and the formation ability of Chinese fir explants.
