目的:建立固相萃取HPLC法测定头孢噻利血药浓度的方法。方法:色谱柱:Hypersil C_(18)(4.6 mm×250 mm,10.0μm),流动相:乙腈-磷酸二氢钾缓冲液(9:91),检测波长:236 nm,流速:0.8 ml·min~(-1)。结果:硫酸头孢噻利在0.09~58.88 mg·L~(-1)范围内呈良好的线性关系(R~2=0.999 7);低、中、高三种浓度的硫酸头孢噻利日内精密度RSD分别为5.86%、1.76%、1.27%,日间精密度RSD分别为6.51%、2.20%、2.95%;在三种浓度的提取回收率分别为102.68%,102.08%,99.64%,RSD值分别为5.19%,3.24%,1.08%。结论:本方法简便、快捷、专属性好,可满足临床药代动力学研究与血药浓度监测的要求。
Objective: To establish a HPLC method for the determination of cefoselide concentration. METHODS: The column was Hypersil C18 (4.6 mm × 250 mm, 10.0 μm). The mobile phase was acetonitrile-potassium dihydrogen phosphate buffer (9:91), the detection wavelength was 236 nm, the flow rate was 0.8 ml · min ~ (-1). Results: Ceftezier sulfate showed a good linearity (R ~ 2 = 0.999 7) in the range of 0.09 ~ 58.88 mg · L ~ (-1). The intra- and inter- Respectively, RSD of 5.86%, 1.76%, 1.27%, day precision RSD were 6.51%, 2.20% and 2.95% respectively. The recoveries of three concentrations were 102.68%, 102.08% and 99.64% 5.19%, 3.24%, 1.08%. Conclusion: The method is simple, rapid and specific, which can meet the requirements of clinical pharmacokinetics and plasma concentration monitoring.
