目的:探讨一种新的获得IgY多克隆抗体的方法和效果。方法:用三种常见的细菌病原体金黄色葡萄球菌(SA)、大肠埃希氏菌(EC)、铜绿假单胞菌(PA)制备复合抗原,并经过灭活后免疫来亨蛋鸡产生IgY多克隆抗体。从免疫后的蛋中提取出IgY多克隆抗体,ELISA法测IgY抗体特异性及浓度滴度。SA、EC、PA三种标准菌株体外抑制实验检测IgY抗体抑菌效果。结果:用SA、EC、PA复合抗原成功地免疫来亨蛋鸡产生IgY多克隆抗体,ELISA法检测抗体浓度滴度为100,000;IgY抗体明显抑制SA、EC、PA三种标准菌株的生长。结论:SA、EC、PA三种细菌复合抗原可以刺激来亨蛋鸡产生高浓度滴度抗体IgY,该抗体能明显抑制SA、EC、PA三种标准菌株的生长。
Objective: To explore a new method and effect of obtaining IgY polyclonal antibody. Methods: The three common bacterial pathogens Staphylococcus aureus (SA), Escherichia coli (EC), Pseudomonas aeruginosa (PA) preparation of composite antigen, and after inactivation immune to Hen egg layer produce IgY Polyclonal antibody. IgY polyclonal antibodies were extracted from the immunized eggs and the IgY antibody specificity and concentration titers were measured by ELISA. Antibacterial activity of IgY antibody was tested in vitro by three kinds of standard strains of SA, EC and PA. Results: The IgY polyclonal antibody was successfully immunized with SA, EC and PA complex antigens. The ELISA titer was 100000. IgY antibody significantly inhibited the growth of three standard strains of SA, EC and PA. CONCLUSION: SA, EC and PA combined with three kinds of bacterial antigens can stimulate IgH to produce IgY with high titer. The antibody can significantly inhibit the growth of three standard strains of SA, EC and PA.
