目的研究中性粒细胞明胶酶相关脂质运载蛋白(NGAL)和肾损伤分子-1(KIM-1)在内毒素(LPS)致大鼠急性肾损伤(AKI)早期的表达变化。方法健康雄性Wistar大鼠32只随机分为4组。正常对照组(NC):经尾静脉注射1ml生理盐水;LPS2、4、6h组:LPS剂量为5mg/kg体重,生理盐水稀释至1ml尾静脉注射,复制AKI模型。造模后在各时间点,ELISA法测大鼠血清和尿液中NGAL的含量及尿液中KIM-1的含量,除蛋白法测定血肌酐(SCr)值并比较各组大鼠光镜下肾脏的病理改变。结果与NC组Scr比较:LPS2h组略升高(P>0.05),LPS4、6h组显著升高(P<0.01);LPS2、4、6h组血清和尿液中NGAL及尿液中KIM-1的水平均明显高于NC组(P<0.01),与LPS2h组相比,LPS4h组血清和尿液中NGAL升高幅度小(P>0.05),而尿液中KIM-1显著性升高(P<0.01);光镜下,NC组肾组织结构正常,LPS2、4、6h组肾组织出现不同程度的病理改变。结论 NGAL和KIM-1在AKI早期即出现升高,可作为AKI的早期诊断指标,并且KIM-1有明显的时间依赖性,可作为病情监测指标。
Objective To investigate the expression changes of neutrophil gelatinase-associated lipocalin (NGAL) and renal injury molecule-1 (KIM-1) in the early stage of acute renal injury (AKI) induced by endotoxin (LPS) in rats. Methods Thirty-two healthy male Wistar rats were randomly divided into 4 groups. Normal control group (NC): 1ml normal saline was injected through the caudal vein; LPS2,4,6h group: LPS dose was 5mg / kg body weight, diluted to 1ml normal saline tail vein injection, copy the AKI model. At each time point after the model was established, the content of NGAL in serum and urine of rats and the content of KIM-1 in urine were measured by ELISA, and the serum creatinine (SCr) Kidney pathological changes. Results Compared with Scr in NC group, LPS2h group slightly increased (P> 0.05), LPS4,6h group increased significantly (P <0.01); LPS2,4,6h group serum and urine NGAL and urine KIM-1 (P <0.01). Compared with LPS2h group, the increase of NGAL in serum and urine of LPS4h group was small (P> 0.05), while the level of KIM-1 in urine was significantly increased P <0.01). Under the light microscope, the structure of kidney in NC group was normal, and the pathological changes of kidney in LPS2, 4, 6h group were different. Conclusions NGAL and KIM-1 are elevated in the early stage of AKI, which can be used as an early diagnosis index of AKI, and KIM-1 has obvious time-dependent and can be used as disease monitoring indicators.
