目的:探讨重组慢病毒介导的基质金属蛋白酶-9(MMP-9)基因沉默对喉鳞状细胞癌移植瘤生长的抑制作用。方法:构建喉癌移植瘤模型,采用RNA干扰技术,瘤内注射MMP-9-RNAi-Lentivirus重组慢病毒,以空病毒载体作为对照,观察抑瘤效果。运用Western-blot方法检测移植瘤中MMP-9蛋白表达。运用免疫组织化学方法检测移植瘤中增殖细胞核抗原(PCNA)的表达,评估其对细胞增殖的影响。结果:重组慢病毒MMP-9-RNAi-Lentivirus治疗后,治疗组裸鼠移植瘤平均重量为(1.484±0.391)g,明显小于对照组(2.618±0.465)g(P<0.05)。治疗组肿瘤平均体积为(1.177±0.270)cm3,明显小于对照组(2.034±0.366)cm3(P<0.05),抑瘤率为43.32%。Western-blot结果显示治疗组MMP-9蛋白表达阴性7例,弱阳性3例,对照组MMP-9蛋白表达均阳性。免疫组织化学显示治疗组PCNA指数为(55.41±8.77)%,显著低于对照组(77.04±6.91)%(P<0.05)。结论:MMP-9-RNAi-Lentivirus重组慢病毒能有效抑制喉癌移植瘤生长及增殖。
Objective: To investigate the inhibitory effect of recombinant lentivirus-mediated matrix metalloproteinase-9 (MMP-9) gene silencing on the growth of laryngeal squamous cell carcinoma. Methods: The model of laryngeal carcinoma xenografts was constructed. RNA interference technique was used to infect intratumoral injection of MMP-9-RNAi-Lentivirus recombinant lentivirus. The empty vector was used as a control to observe the anti-tumor effect. Western-blot method was used to detect the expression of MMP-9 in xenografts. Immunohistochemistry was used to detect the expression of proliferating cell nuclear antigen (PCNA) in the transplanted tumor and evaluate its effect on cell proliferation. Results: After treatment with recombinant lentivirus MMP-9-RNAi-Lentivirus, the average weight of transplanted tumor in nude mice in treatment group was (1.484 ± 0.391) g, which was significantly lower than that in control group (2.618 ± 0.465) g (P <0.05). The mean tumor volume in the treatment group was (1.177 ± 0.270) cm 3, which was significantly lower than that in the control group (2.034 ± 0.366 cm 3) (P <0.05). The tumor inhibition rate was 43.32%. Western-blot results showed that the expression of MMP-9 in the treatment group was negative in 7 cases, weakly positive in 3 cases, and that in the control group was positive in MMP-9 protein. Immunohistochemistry showed that the PCNA index in the treatment group was (55.41 ± 8.77)%, which was significantly lower than that in the control group (77.04 ± 6.91)% (P <0.05). Conclusion: The recombinant lentivirus of MMP-9-RNAi-Lentivirus can effectively inhibit the growth and proliferation of laryngeal carcinoma.
