目的:探讨黄芩素对单侧输尿管结扎致大鼠肾间质纤维化的干预作用及机制。方法:将40只雄性SD大鼠随机分为假手术组、模型组、科素亚(阳性药)治疗组20 mg/(kg.d)、黄芩素高剂量组200 mg/(kg.d)、黄芩素低剂量组100 mg/(kg.d)。采用单侧输尿管结扎手术,术后10 d处死大鼠。观察肾功能的变化,测定肾组织中羟脯氨酸含量,HE和天狼星红染色光镜下观察肾组织的病理变化,免疫组化法检测肾组织TGF-β1和Smad-2的表达。结果:与假手术组比较,模型组大鼠血肌酐、尿素氮和羟脯氨酸含量显著升高;光镜下发现模型组出现明显的纤维化;模型组较假手术组TGF-β1和Smad-2表达显著升高;各给药组较模型组纤维化程度有所降低。结论:黄芩素可改善肾脏损伤,对肾脏有一定保护作用。
Objective: To investigate the effect and mechanism of baicalein on renal interstitial fibrosis in rats with unilateral ureteral obstruction. Methods: Forty male Sprague-Dawley rats were randomly divided into sham-operated group, model group, coxsackievir (positive drug) treatment group 20 mg / (kg.d), high dose of baicalein group 200 mg / (kg.d) , Baicalein low dose group 100 mg / (kg.d). Unilateral ureteral ligation was performed and rats were sacrificed 10 days after operation. The changes of renal function were observed. The contents of hydroxyproline in renal tissue were measured. The pathological changes of renal tissues were observed under HE and Sirius red staining. The expressions of TGF-β1 and Smad-2 were detected by immunohistochemistry. Results: Compared with the sham-operation group, the content of serum creatinine, urea nitrogen and hydroxyproline in the model group increased significantly. The model group showed obvious fibrosis under the light microscope. Compared with the sham operation group, TGF-β1 and Smad -2 expression was significantly increased; each group than in the model group fibrosis decreased. Conclusion: Baicalein can improve kidney injury and protect the kidneys.
