目的测定不同产地马兜铃药材中马兜铃总酸的含量,为马兜铃总酸限量标准的制定提供必要的数据支持。方法以马兜铃总酸含量为指标,采用L9(34)正交试验方法考察提取溶剂、溶剂用量和提取方法 3个因素,对提取条件进行优化,用紫外分光光度法检测马兜铃总酸的含量,检测波长为319 nm。结果马兜铃总酸最优的提取条件为用含1%甲酸的甲醇,料液比1∶40,超声提取30 min,提取2次;马兜铃酸Ⅰ浓度在0.005~0.030 mg·ml-1与吸光度呈良好的线性关系,A=38.046C+0.0109,r=0.9999,平均回收率为97.79%,RSD=2.09%;不同产地马兜铃种子的马兜铃总酸含量为0.357%~0.597%,果皮的马兜铃总酸含量为0.170%~0.258%。结论不同产地马兜铃药材中马兜铃总酸含量存在明显的差异,并且种子中马兜铃总酸含量明显高于果皮。
Objective To determine the content of aristolochic acid in aristolochia officinalis from different areas and provide necessary data support for the establishment of aristolochic acid total acid limit standard. Methods Taking total aristolochic acid content as an index, the factors of extraction solvent, solvent dosage and extraction method were investigated by L9 (34) orthogonal test. The extraction conditions were optimized. The contents of aristolochic acid The detection wavelength is 319 nm. Results The optimum extraction conditions of total acid were as follows: extraction with methanol containing 1% formic acid at a solid-to-liquid ratio of 1:40 and ultrasonic wave extraction for 30 min, twice with aristolochic acid at a concentration of 0.005-0.030 mg · ml- 1 showed a good linear relationship with the absorbance. The average recovery was 97.79% and the RSD was 2.09% for A = 38.046C + 0.0109 and r = 0.9999, respectively. The total contents of aristolochic acid in Aristolochia species were 0.357% ~ 0.597 %, Aristolochic total acid content of aristolochic acid 0.170% ~ 0.258%. Conclusion The total acid content of aristolochiae in the different producing areas has obvious difference, and the total aristolochic acid content in the seeds is obviously higher than that in the pericarp.
