作者用半定量逆转录酶-多聚酶链反应(RT-PCR)方法,分析了自体骨髓(BM)和生长因子动员的外周血于细胞(PSC)产品中单个核细胞的核型、功能及稳定状态下细胞因子基因的表达,并与正常外周血单个核细胞(PBMC)及植物血凝素(PHA)激活的PBMC进行比较。结果:(1)PSC产品中CD3、CD4及CD8阳性细胞显著高于BM产品(P≤0.01),而与PBMC类似。CD14阳性的单核细胞在PSC中含量远高于BM及PBMC(P≤0.001);(2)PSC产品中单个核细胞对PHA促分裂反应指数显著高于BM(P≤0.05),而PSC及BM该指数均显著低于PBMC(P≤0.05)。这提示干细胞产品免疫功能受抑制;(3)PSC产品中IL-2基因表达水平显著高于BM产品(P=0.006),而PSC
The authors used semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) to analyze the karyotype, function, and stability of mononuclear cells in autologous bone marrow (BM) and peripheral blood mobilized from growth factors The expression of cytokines was compared with that of normal peripheral blood mononuclear cells (PBMCs) and phytohaemagglutinin (PHA) -activated PBMCs. Results: (1) CD3, CD4 and CD8 positive cells in PSC products were significantly higher than BM products (P≤0.01), but similar to PBMCs. CD14 positive monocytes were much higher in PSC than those in BM and PBMC (P≤0.001).
