参麦注射液对环孢霉素A肾毒性的拮抗作用及机制探讨

目的探讨参麦注射液预防环孢霉素A中毒性肾损伤的机制。方法实验大鼠分对照组、环孢霉素A中毒组及参麦注射液处理组,分别给予橄榄油灌胃,环孢霉素A灌胃,环孢霉素A灌胃同时参麦注射液腹腔注射,连续4周后,取肾组织作病理观察,并测肾组织超氧化物歧化酶和丙二醛含量,取血测血肌酐、尿素氮和血流变学指标。结果与对照组相比,环孢霉素A组大鼠,肾脏功能明显受损,血肌酐、尿素氮明显增高;血液呈高黏滞状态,全血黏度和红细胞压积显著降低,血浆黏度,红细胞聚集指数,纤维蛋白原显著增高;肾组织氧化性损伤加重,超氧化物歧化酶含量减低而氧化产物丙二醛MDA含量增高;与环孢霉素A中毒组相比,参麦注射液组大鼠肾脏功能受损明显减轻,血肌酐、尿素氮降低,脂质过氧化损伤减轻,血流变各项指标均有明显改善。结论参麦注射液通过抗脂质过氧化,减轻血流变障碍从而减轻环孢霉素A引起的中毒性肾损伤。 Objective To explore the mechanism of Shenmai injection in preventing renal injury induced by cyclosporine A. Methods The experimental rats were divided into control group, cyclosporine A poisoning group and Shenmai injection group, respectively, with olive oil orally, cyclosporin A gavage, gavage with CsA injection The rats were injected intraperitoneally for 4 weeks. The pathological changes of renal tissues were observed. The content of superoxide dismutase and malondialdehyde in renal tissues was measured. Blood samples were collected for serum creatinine, urea nitrogen and hemorheology. Results Compared with the control group, the function of kidney in cyclosporin A group was significantly impaired, serum creatinine and urea nitrogen were significantly increased; the blood was viscous, the whole blood viscosity and hematocrit were significantly decreased, the plasma viscosity, Erythrocyte aggregation index and fibrinogen were significantly increased; oxidative damage of renal tissue was increased, the content of superoxide dismutase was decreased and MDA content of oxidized product was increased; Compared with cyclosporin A poisoning group, Shenmai injection group Rat renal function was significantly reduced, serum creatinine, urea nitrogen decreased, lipid peroxidation lesion reduced, blood rheology indicators were significantly improved. Conclusion Shenmai injection can alleviate the toxic renal injury induced by cyclosporine A through anti-lipid peroxidation and reduce the dysfunction of blood rheology.
