ECE支TCP基因调控植物的分枝和花的对称性,但其功能在不同物种间存在差异。利用简并PCR法结合RACE技术从矮牵牛(Petunia hybrida Vilm)GL8自交系中获得了4个矮牵牛ECE支TCP基因家族成员的全长cDNA序列,分别命名为PhTCP1~4(登录号:JQ400104~JQ400107)。cDNA和基因组序列分析表明,PhTCP1~4分别编码406、332、341和343个氨基酸,PhTCP1不含内含子,其余3个基因含1~2个内含子。进化树分析发现,PhTCP1~4分属于CYC1、CYC2和CYC3分支。荧光定量PCR分析表明,PhTCP1~4均在成株期矮牵牛腋芽中优势表达,在不同节位腋芽中表现的表达趋势各不相同,提示矮牵牛ECE支TCP基因可能主要与腋芽的生长发育相关。
The ECE-TCP gene regulates plant branching and flower symmetry, but its function varies among species. The full-length cDNA sequences of four members of the ECE-TCP gene family from Petunia hybrida Vilm GL8 were obtained by degenerate PCR and RACE techniques, and named as PhTCP1-4 (accession number : JQ400104 ~ JQ400107). cDNA and genome sequence analysis showed that PhTCP1 ~ 4 encoded 406,332,341 and 343 amino acids, PhTCP1 did not contain introns, the remaining three genes containing 1 or 2 introns. Phylogenetic tree analysis found that PhTCP 1 ~ 4 belong to CYC1, CYC2 and CYC3 branches. Fluorescent quantitative PCR analysis showed that PhTCP1 ~ 4 were predominantly expressed in axillary buds of adult petunia, and expressed differently in axillary buds at different node stages, suggesting that the TCP gene of ECE in petunia may be mainly related to axillary bud growth Developmental related.
