目的比较4种血清型登革病毒NS1蛋白型特异性抗原表位基因序列及氨基酸序列之间的差异,为利用基因差异进行血清学分型及疫苗研究提供新的线索。方法利用DNAstar数据包中的Editseq程序,从20株登革病毒分离株的全基因组序列中将NS1基因型特异性抗原表位序列截取出来,再用ClustalX软件进行多序列比对,进行同源性分析,找出型内最为保守的抗原表位序列。并将比对结果在120株登革病毒序列中进一步验证。结果 NS1蛋白36~45和71~85位氨基酸为型特异性抗原表位,高度保守,型内完全相同,型间不同。结论 NS1蛋白36~45位氨基酸可以作为登革病毒血清分型和研制亚单位疫苗的靶标。
Objective To compare the differences in the sequence and amino acid sequence of NS1 epitope of four serotypes of dengue virus, and to provide new clues for serological typing and vaccine research using genetic differences. Methods Using the Editseq program in the DNAstar package, the NS1 genotype-specific epitopes were cloned from the genome-wide sequences of 20 dengue virus isolates. Multiple sequence alignments were performed using ClustalX software, and homology Analysis, find the type of the most conserved epitope sequence. The results of the comparison were further validated in 120 dengue virus sequences. Results The amino acids 36-45 and 71-85 of NS1 protein were type-specific antigen epitopes, highly conserved, completely identical in type, and different in type. Conclusion The amino acids 36-45 of NS1 protein can be used as the target of dengue virus serotyping and subunit vaccine development.
