目的 利用人肺鳞移植瘤模型研究平肺口服液的抗瘤作用机理。方法 采用人肺鳞裸鼠移植模型 ,分组分别给予生理盐水、平肺口服液及顺铂。给药 2 0d后无菌取瘤组织 ,观察肿瘤大小、肿瘤细胞增殖状况、肿瘤血管密度、细胞内皮生长因子 (VEGF)的蛋白表达。结果 平肺口服液能明显抑制肿瘤细胞增殖 ,但不及化疗组 ,化疗组肿瘤血管和模型组比有所减少 ,但不如平肺组减少明显 ,化疗组血管密度高于平肺组 ,平肺组的阳性细胞面积和平均视觉密度都明显少于模型组 ,化疗组VEGF阳性细胞染色深度有所增加。结论 以养阴清肺法为治则的平肺口服液对肺癌有明显的治疗作用 ,本研究初步证实抑制细胞增殖和影响肿瘤新生血管形成是其作用机理之一。
Objective To study the mechanism of anti-tumor effect of Pingfei oral liquid by using human lung squamous tumor xenograft model. Methods Human lung squamous nude mice transplantation model was used to administer physiological saline, Pingfei oral liquid and cisplatin. After 20 days of administration, tumor tissues were aseptically removed to observe tumor size, tumor cell proliferation, tumor vascular density, and protein expression of vascular endothelial growth factor (VEGF). Results Pingfei oral liquid could obviously inhibit the proliferation of tumor cells, but it was less than chemotherapy group. The ratio of tumor blood vessels and model group in chemotherapy group was decreased, but it was not as obvious as that in Pingfei group. The vascular density in chemotherapy group was higher than that in Pingfei group and positive in Pingfei group. Cell area and average visual density were significantly lower than those in the model group. The staining depth of VEGF-positive cells in the chemotherapy group increased. Conclusion Pingyin Oral Liquid with Nourishing Yin and clearing the lung is a significant therapeutic effect on lung cancer. This study has initially demonstrated that inhibiting cell proliferation and influencing tumor neovascularization is one of its mechanisms.
