目的探讨慢性氟中毒对雄性大鼠睾丸组织某些生化指标水平的影响,为氟的生殖毒性研究提供实验依据。方法健康雄性Wistar大鼠32只,体重150~180 g,随机分为4组:对照组、低、中和高氟组[100、200和300 mg/(kg·d)NaF],灌胃染毒90 d,每天称体重。染毒结束后处死大鼠,摘取睾丸组织,称重并计算脏器系数;分光光度法检测睾丸组织匀浆中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及一氧化氮合酶(NOS)的活性和丙二醛(MDA)、过氧化氢(H2O2)及一氧化氮(NO)的含量。结果染氟第30天大鼠体重组间比较,差异有统计学意义(P<0.05),其中低、中氟组高于高氟组(P<0.05);第0、60和90天大鼠体重组间比较,差异无统计学意义(P>0.05)。与对照组比较,各组睾丸组织的SOD和CAT活性无显著性改变(P>0.05),低氟组MDA、中氟组H2O2含量均有显著性升高(P<0.05),低氟组NOS活性显著性降低(P<0.01)。结论慢性氟中毒可增强睾丸组织氧化应激效应,损害大鼠生殖系统。
Objective To investigate the effect of chronic fluorosis on some biochemical indexes in testis of male rats and provide experimental basis for the study of reproductive toxicity of fluorine. Methods Thirty-two healthy male Wistar rats weighing 150-180 g were randomly divided into 4 groups: control group, low, medium and high fluoride group [100, 200 and 300 mg / (kg · d) NaF] Toxic 90 d, weighing each day. The rats were sacrificed at the end of the exposure and the testicular tissues were harvested and weighed and the organ coefficients calculated. The contents of superoxide dismutase (SOD), catalase (CAT) and nitric oxide Enzyme (NOS) activity and the content of malondialdehyde (MDA), hydrogen peroxide (H2O2) and nitric oxide (NO). Results Compared with the high fluoride group (P <0.05), the body weight of the rats on the 30th day after fluorine exposure were significantly different (P <0.05) There was no significant difference between weight groups (P> 0.05). Compared with the control group, there was no significant difference in the activity of SOD and CAT in testis (P> 0.05), the content of H2O2 in MDA and Fluoride groups were significantly increased in low fluoride group (P <0.05) Activity was significantly lower (P <0.01). Conclusion Chronic fluorosis can enhance the oxidative stress in testis tissue and impair the reproductive system in rats.
