为探讨一氧化氮及抗氧化酶在石棉致病中的作用 ,用改良 Myrvik法收集兔肺泡巨噬细胞 (AM)进行体外培养 ,采用硝酸还原酶法测定 U ICC温石棉处理后兔 AM培养液中亚硝酸根 (NO2 - )的含量 (NO的静态氧化形式 ) ,同时利用黄嘌呤及黄嘌呤氧化酶反应系统测定 AM的细胞裂解液中 SOD的活性 ,和酶 -底物反应形成有色物质来测定 AM的细胞裂解液中 NOS和 GSH - Px的酶活性。结果显示 :随 UICC温石棉剂量的增加出现以下情况 :1 AM死亡率升高 ,细胞活性下降 ;2 AM培养液中的 NO- 2 含量增加 ,AM细胞裂解液中 SOD和 GSH - Px的活性降低 ,且以上均有剂量 -效应关系 (r1 =0 .6 1 1 4 ,P<0 .0 5 ;r2 =- 0 .5 1 2 6 ,P<0 .0 1 ;r3=- 0 .91 37,P<0 .0 1 ) ;3NOS在较低剂量组间呈剂量 -反应性增加 ,而在较高剂量组间呈剂量 -反应性降低 ,出现一个单峰曲线 ;4测得的NO的含量分别与 SOD和 GSH- Px活性呈负相关关系 (r1 =- 0 .71 2 5 ,P<0 .0 5 ;r2 =- 0 .8496 ,P<0 .0 5 ) ;5 NO和NOS在较低剂量组间呈正相关关系 ,在较高剂量组间呈负相关关系。以上提示 NO及其抗氧化酶在温石棉致病中具有一定作用
