为了研究胰岛素样生长因子-1受体在血管平滑肌细胞生长调节中所起的作用,用其寡核苷酸反义链和配链对鼠主动脉平滑肌细胞的作用进行研究。用寡核苷酸反义链处理的血管平滑肌细胞中,氚标脱氧胸腺嘧啶核苷的掺入率较对照组下降64%,用结合实验测定胰岛素样生长因子-1受体数目下降52%。在用反义链处理细胞时,随加入胰岛素样生长因子-1浓度的增加,氚标脱氧胸腺嘧啶核苷掺入率也与剂量成反比的下降。反义键的剂量为20μg/L时,氚标脱氧胸腺嘧啶核苷掺入率下降64%。反义链处理的细胞对加入血管紧张素Ⅱ无反应。受体数目无变化。这一发现提示了配体一受体系统对血管平滑肌细胞反应的重要性。受体的密度对血管平滑肌细胞的生长起到关键作用。用反义链能使胰岛素生长因子-1受体数目减少是由于胰岛素样生长因子-1受体的mRNA减少,这一假说在核酸酶保护实验中得到证明。
In order to investigate the role of insulin-like growth factor-1 receptor in the regulation of vascular smooth muscle cell growth, the effect of its antisense oligonucleotides and its strands on mouse aortic smooth muscle cells was studied. The incorporation of tritiated thymidine into vascular smooth muscle cells treated with the antisense oligonucleotide oligonucleotide decreased by 64% compared with the control group, and the number of insulin-like growth factor-1 receptors decreased by 52% as measured by the binding assay. When cells were treated with an antisense strand, the incorporation of tritiated thymidine also decreased inversely with dose as the concentration of insulin-like growth factor-1 was added. Antisense bond dose of 20μg / L, tritiated thymidine incorporation decreased by 64%. Antisense-treated cells did not respond to addition of angiotensin II. No change in the number of receptors. This finding suggests the importance of the ligand-receptor system for vascular smooth muscle cell responses. The density of receptors plays a key role in the growth of vascular smooth muscle cells. The reduction of the number of insulin-like growth factor-1 receptors with the antisense strand is due to a decrease in the mRNA of the insulin-like growth factor-1 receptor, a hypothesis that has been demonstrated in nuclease protection experiments.
