以华盖木4年生嫁接苗当年生枝为外植体,在初代培养过程中每周测定外植体的多酚氧化酶(PPO)活性及总酚含量,并统计外植体的褐化率,对华盖木初代培养过程中外植体的褐化与PPO活性及酚类物质含量的关系进行分析。结果表明:华盖木外植体的总酚含量于接种后第2周时达到最大值2.4445mg/g。此后又快速下降。接种后1周内酶活性快速增加,第1~3周,酶活性又大幅度下降,第3~4周酶活性下降速度减缓。接种后第2、3周是褐化的高发生期,第3周后褐化率的增长较慢。外植体的褐化受着PPO活性和酚类物质含量的共同影响。
The branches of four-year-old grafted seedlings were used as explants. The activity of polyphenol oxidase (PPO) and total phenolic content of explants were measured weekly during the primary culture. The explants’ The relationship between browning and PPO activity and phenolic content of explants during the primary culture of Culter alburnii was analyzed. The results showed that the total phenolic content of the explants reached the maximum of 2.4445mg / g at the second week after inoculation. After that, it dropped rapidly again. Within 1 week after inoculation, the enzyme activity rapidly increased. In the first to third weeks, the enzyme activity dropped significantly and the rate of decrease of enzyme activity in the third to the fourth week slowed down. Weeks 2 and 3 of inoculation were the high stage of browning, and the rate of browning increased slowly after 3 weeks. The browning of explants is influenced by PPO activity and phenolic content.
