利用根癌农杆菌(Agrobacteriumtumefacicns)EHA101携带反义PEP基因和卡那霉素、潮霉素抗性基因,采用共培养法转化甘蓝型油菜(BrassicanapusL.)浙油758和浙优油1号的下胚轴,通过组织培养获得了能在潮霉素培养基上生长的油菜转基因植株。经GUS组织化学染色及PCR扩增检测,证明外源基因已插入油菜基因组并得到表达。浙油758转基因植株T1代种子平均含油量比对照提高6.4%,其中一株比对照高15.15%;浙优油1号转基因植株T1代种子平均含油量比对照提高16.7%,有多株比对照提高18%以上,最高含油量达49.54%,蛋白质含量与油脂含量呈显著负相关。
Using Agrobacterium tumefaciens EHA101 to carry antisense PEP gene and kanamycin and hygromycin resistance genes, co-culture method was used to transform Brassica napus L.Zheyou 758 and Zheyouyou 1 Hypocotyledonous embryos, rapeseed transgenic plants that can grow on hygromycin medium were obtained by tissue culture. The GUS histochemical staining and PCR amplification tests showed that exogenous genes have been inserted into the genome of rape and expressed. The average seed oil content of T1 generation of Zheyou 758 transgenic plants increased by 6.4% compared with the control, one of them was 15.15% higher than that of the control. The average oil content of T1 seeds of Zhejiang Youyou 1 transgenic plants was increased by 16.7% , More than 18% higher than the control, the highest oil content of 49.54%, protein content and oil content was significantly negatively correlated.
