目的:观察纳米银杏(银杏黄酮-磷脂复合物)对动脉粥样硬化(AS)早期大鼠主动脉壁细胞间黏附分子-1(ICAM-1)mRNA及其蛋白表达水平的影响。方法:采用生化分析、免疫组化、实时定量荧光RT-PCR以及凝胶电泳,观察纳米银杏对大鼠主动脉ICAM-1mRNA及其蛋白表达水平的影响。结果:与模型组比较,纳米银杏组ICAM-1表达明显降低(P<0.01);与银杏黄酮组比较,纳米银杏组ICAM-1表达明显降低(P<0.01)。结论:银杏黄酮及纳米银杏可下调AS早期大鼠主动脉壁ICAM-1的高表达,纳米银杏的作用明显优于银杏黄酮。
Objective: To observe the effect of nano-Ginkgo biloba (Ginkgo biloba flavonoids-phospholipid complex) on the expression of intercellular adhesion molecule-1 (ICAM-1) mRNA and protein in the aorta of rats with atherosclerosis (AS). Methods: Biochemical analysis, immunohistochemistry, real-time quantitative RT-PCR and gel electrophoresis were used to observe the effect of nano-Ginkgo biloba on the expression of ICAM-1 mRNA and protein in rat aorta. RESULTS: Compared with the model group, the expression of ICAM-1 in the Ginkgo biloba group was significantly lower (P<0.01). Compared with the ginkgo flavonoid group, the expression of ICAM-1 in the Ginkgo biloba group was significantly lower (P<0.01). CONCLUSION: Ginkgo biloba flavonoids and nano-ginkgo biloba can down-regulate the expression of ICAM-1 in the aortic wall of rats with early AS. The effect of nano-ginkgo biloba is significantly better than that of ginkgo flavone.
