目的观察黄粉虫抗菌肽对K562细胞周期的影响;比较该抗菌肽与羟基脲对K562细胞的增殖抑制作用。方法用流式细胞仪检测黄粉虫抗菌肽对K562细胞周期的影响。黄粉虫抗菌肽与羟基脲分别设置5个浓度组,用MTT比色法分别求出2种药物不同浓度对K562的抑制率,根据浓度和抑制率用SPSS软件求出回归方程和IC50。结果细胞周期G0/G1期和G2/M期细胞比例增加,S期细胞比例下降。其中只有G0/G1期和S期与对照组相比,差异具有统计学意义(P<0.01)。说明抗菌肽可使细胞周期阻滞于G0/G1期,从而抑制S期DNA的合成。抗菌肽和羟基脲的IC50分别为29.98和3644.45μg/ml。结论抗菌肽可通过影响细胞周期来发挥抗K562细胞的作用。抗菌肽明显优于羟基脲对K562细胞的增殖抑制作用,有较好的应用前景。
Objective To observe the effect of antibacterial peptides of Tenebrio molitor on the cell cycle of K562 cells and compare the inhibitory effects of the antibacterial peptides and hydroxyurea on the proliferation of K562 cells. Methods Flow cytometry was used to detect the effect of Tenebrio molitor peptides on the cell cycle of K562 cells. Tenebrio molitor peptides and hydroxyurea were set at 5 concentration groups. MTT colorimetric method was used to determine the inhibitory rates of K562 at different concentrations. The regression equation and IC50 were obtained by SPSS software according to the concentration and inhibition rate. Results The proportion of cells in G0 / G1 phase and G2 / M phase increased and the percentage of S phase cells decreased. Only G0 / G1 phase and S phase compared with the control group, the difference was statistically significant (P <0.01). Antibacterial peptides can block the cell cycle in the G0 / G1 phase, thereby inhibiting the synthesis of S-phase DNA. The IC50 of antimicrobial peptide and hydroxyurea were 29.98 and 3644.45μg / ml, respectively. Conclusion Antimicrobial peptides exert the anti-K562 effect by affecting the cell cycle. Antibacterial peptides were significantly better than hydroxyurea on K562 cell proliferation inhibition, has a good prospect.
