目的:通过对8份夏枯草种质资源的AFLP分析,探求各种质间的遗传多样性。方法:采用扩增片段长度多态性(AFLP)标记,在DNA水平上进行遗传多样性研究,筛选了32对选择性扩增引物,将扩增出的条带作为原始矩阵,用NTSYS-PC软件计算并分析了夏枯草种质间的相似度,构建了遗传系统进化树。结果:①SDS法提取的夏枯草基因组DNA质量较佳,能够满足AFLP分析的要求;②从32对选择性扩增引物中,筛选出10对多态性较强、带型较好、分辨率较高的组合;③构建了夏枯草AFLP指纹图谱,将8个夏枯草种质全部区分开来;④通过构建进化树,把8个种质分成3类。结论:夏枯草种质资源有丰富的遗传多样性,某些形态特征差异和基因型存在相关性。
OBJECTIVE: To explore the genetic diversity of various interspecies by AFLP analysis of 8 Prunella vulgaris germplasm resources. Methods: Amplified fragment length polymorphism (AFLP) markers were used to study the genetic diversity at the DNA level. Thirty-two pairs of selective amplification primers were screened and the amplified bands were used as the original matrix. NTSYS-PC was used. The software calculated and analyzed the similarity between the Prunella vulgaris germplasm and constructed a genetic phylogenetic tree. RESULTS: 1The genomic DNA of Prunella vulgaris extracted by SDS method was better, which could meet the requirements of AFLP analysis.2 From 10 pairs of selective amplification primers, 10 pairs of selective amplification primers were screened out. High combination; 3 Prunella AFLP fingerprinting was constructed and all 8 Prunella spp. germplasms were separated; 4 Phylogenetic trees were used to classify 8 germplasms into 3 categories. Conclusion: Prunella vulgaris germplasm resources have abundant genetic diversity, and there are correlations between some morphological characteristics and genotypes.
