More and more evidences indicate that the method of mouse footpad model needs to be modified in order to determine the drug resistance of M. leprae, to monitor the chemotherapy and to screen for new anti-leprosy drugs, so that the activity of M. leprae can be determined early. Several in vitro assays have been reported over the past decade. These include the incorporation of radiolabels into M. leprae grown in macrophages or special media, determination of ATP levels, determination of esterase and intracellular cation ratios, etc. Labs have routinely used M. leprae cultured in macrophages for ~ 3H-thymidine incorporation assays as DNA fingerprinting for M. leprae
