目的建立高效液相色谱法测定大腹皮中槟榔碱的含量。方法采用Thermo-BioBasic SCX离子交换柱(4.6 mm×250mm,5μm),流动相为乙腈-0.2%磷酸溶液(55∶45),浓氨试液调节pH值至3.8,流速1.0 mL.min-1,柱温30℃,检测波长215 nm。结果氢溴酸槟榔碱的色谱峰面积与质量浓度呈良好的线性关系,线性范围为5.2～83.2μg.mL-1(r=0.999 1),平均回收率(n=9)为98.8%。结论该方法简便、准确、专属性强,为大腹皮饮片的质量评价提供了方法。
Objective To establish a HPLC method for the determination of arecoline in the large belly skin. Methods A Thermo-BioBasic SCX column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase was acetonitrile-0.2% phosphoric acid solution (55:45). The concentrated ammonia solution was adjusted to pH 3.8 and the flow rate was 1.0 mL min -1 , Column temperature 30 ℃, detection wavelength 215 nm. Results The arealine hydrobromide peak area showed a good linear relationship with the mass concentration. The linear range was from 5.2 to 83.2μg.mL-1 (r = 0.999 1). The average recoveries (n = 9) were 98.8%. Conclusion The method is simple, accurate and specific, which provides a method for the quality evaluation of the large belly skin Pieces.
