1974年中国医学科学院药物研究所与四川抗菌素工业研究所分别从广东省大埔县和四川省南川县土壤中分离到链霉菌1748和10204,两株链霉菌经培养后其产物的理化特性及生物活性鉴定证明与国外的麦迪霉素(Mydecamycin)相同。麦迪霉素是含多组份的抗生素,我们参照Omura,S.等所报道的高效液相色谱分离大环内酯类抗生素的方法,用岛津高效液相色谱仪LC-3A,分离柱Zorbax ODS(25cm×4.6mm),柱温恒定,柱压160kg/cm~2,甲醇:W/15醋酸盐缓冲液(pH4.9)(6∶4)为流动相,样品以流动相溶解,浓度为1.5mg/ml,滤膜过滤后,每次进样10~15μl,流速1.0ml/min,波长232.5nm检测,测定的样品有日本商品和国内的一些标准品和有关生产厂的某些批号产品,测定结果见图1~图7及表一。
Streptomyces 1748 and 10204 were isolated from the soil of Dapu County of Guangdong Province and Nanchuan County of Sichuan Province in 1974 respectively by Institute of Materia Medica, Chinese Academy of Medical Sciences and Sichuan Institute of Antibiotics. The physical and chemical properties and biological The activity identification proved to be the same as Mydecamycin abroad. Miconicycin is a multi-component antibiotic, we refer to Omura, S. and other reported high-performance liquid chromatography macrolide antibiotics, with Shimadzu high performance liquid chromatography LC-3A, column Zorbax ODS (25cm × 4.6mm), constant column temperature, column pressure 160kg / cm ~ 2, methanol: W / 15 acetate buffer pH4.9 (6: 4) as the mobile phase. Concentration of 1.5mg / ml, the membrane filter, each injection of 10 ~ 15μl, the flow rate of 1.0ml / min, wavelength 232.5nm detection, the determination of the sample of Japanese and some domestic products and some of the relevant standard Lot number products, the results shown in Figure 1 ~ Figure 7 and Table I.
