目的通过体内模拟腹腔镜气腹环境,探讨CO2气腹对裸鼠腹腔胃癌细胞增殖和侵袭转移能力的影响。方法裸鼠随机分成3组:对照组(A组),行开腹探查;5mmHg(1mmHg=0.133kPa)CO2气腹组(B组);8mmHgCO2气腹组(C组);每组9只。人胃癌细胞SGC-7901裸鼠腹腔注射后建立CO2气腹胃癌细胞裸鼠腹腔种植模型,气腹持续1h。建模后每周秤体质量,观察各组裸鼠生长情况和腹腔成瘤情况。28d后处死解剖裸鼠,记录腹水量、腹腔各脏器、穿刺口和腹壁切口成瘤情况和肿瘤数,测量肿瘤最大径、总重量,并检测胃癌种植瘤组织中乙酰肝素酶(Hpa)mRNA的表达。结果各组裸鼠成瘤率100%。各组裸鼠体质量、腹腔不同脏器成瘤率、腹水量差异均无统计学意义(P>0.05)。肿瘤结节数:A组(7.4±2.5)个,B组(7.3±2.4)个,C组(7.5±2.1)个。肿瘤最大径:A组(7.0±2.2)mm,B组(6.9±2.0)mm,C组(6.8±1.9)mm;肿瘤重量:A组(0.99±0.30)g,B组(0.87±0.20)g,C组(0.91±0.13)g。HpamRNA表达指数:A组0.41±0.07,B组0.45±0.06,C组0.43±0.09,各组间差异均无统计学意义(P>0.05)。结论在5mmHg和8mmHg气腹压力下,CO2气腹不促进胃癌细胞裸鼠腹腔内增殖和侵袭转移,也未明显影响胃癌细胞成瘤组织HpamRNA的表达。
Objective To study the effects of CO2 pneumoperitoneum on the proliferation, invasion and metastasis of abdominal cancer cells in nude mice by simulating laparoscopic pneumoperitoneum in vivo. Methods Nude mice were randomly divided into 3 groups: control group (group A), open laparotomy; 5mmHg (1mmHg = 0.133kPa) CO2 pneumoperitoneum group (B group); 8mmHgCO2 pneumoperitoneum group (C group); Gastric cancer cells SGC-7901 nude mice after intraperitoneal injection of CO2 pneumoperitoneum to establish a model of intraperitoneal implantation of pneumoperitoneum, pneumoperitoneum for 1h. After modeling the body mass every week, observe the growth of nude mice in each group and the status of celiac tumorigenesis. After 28 days, the nude mice were sacrificed and the ascites volume, intraperitoneal organs, puncture and abdominal incision tumor formation and tumor number were recorded. The maximum diameter and total weight of the tumors were measured. The heparanase (Hpa) mRNA expression. Results The tumor formation rate of nude mice in each group was 100%. The body weight of nude mice in each group, tumor formation rate of different organs in the abdominal cavity, ascites volume difference was not statistically significant (P> 0.05). The number of tumor nodules was 7.4 ± 2.5 in group A, 7.3 ± 2.4 in group B and 7.5 ± 2.1 in group C, respectively. The maximum diameter of tumor in group A was (7.0 ± 2.2) mm, in group B (6.9 ± 2.0) mm, in group C (6.8 ± 1.9) mm. The tumor weight in group A was 0.99 ± 0.30 g and in group B was 0.87 ± 0.20, g, C group (0.91 ± 0.13) g. HpamRNA expression index: A group of 0.41 ± 0.07, B group 0.45 ± 0.06, C group 0.43 ± 0.09, no significant difference between the groups (P> 0.05). Conclusions CO2 pneumoperitoneum did not promote intraperitoneal proliferation, invasion and metastasis of gastric cancer cells in nude mice at 5 mmHg and 8 mmHg pneumoperitoneum pressure, nor did it significantly affect the expression of HpamRNA in gastric cancer cells.
