目的:检测新型有机硒化合物双硒唑烷-1(Eb)的动物体内抗肿瘤作用。方法:建立Lewis肺癌(lewislungcancer,LLC)皮下移植瘤C57/BL鼠动物模型,选取25.0mg/kg和12.5mg/kg两个剂量的Eb作为实验组,以2.0mg/kg剂量的顺铂(DDP)作为阳性对照,以溶剂5g/L羧甲基纤维素钠溶液为阴性对照,于接种肿瘤后第2天开始向C57/BL鼠腹腔连续注射药物7d,探讨Eb对荷瘤鼠的存活期、肿瘤的生长速度、浸润级别、细胞形态、细胞周期和细胞凋亡的影响。结果:Eb能够明显抑制肿瘤生长和侵袭(高剂量Eb的肿瘤抑制率为80.31%),延长荷瘤鼠的存活期;形态学观察发现,给予Eb后肿瘤细胞核浓缩深染,分裂相细胞减少;流式细胞仪检测结果表明,Eb能够促进肿瘤细胞的凋亡。结论:新型有机硒化合物Eb在C57/BL小鼠体内能够明显抑制LLC的生长和侵袭,促进肿瘤细胞的凋亡,具有较强的抗肿瘤活性。
OBJECTIVE: To test the anti-tumor effect of a novel organic selenium compound bisselenazolidine-1 (Eb) in animals. Methods: Animal models of Lewis lung carcinoma (LLC) C57 / BL mice were established. Eb of 25.0 mg / kg and 12.5 mg / kg were selected as the experimental group and DDP (2.0 mg / kg) ) As a positive control, the solvent 5g / L sodium carboxymethyl cellulose solution as a negative control, starting from 2 days after tumor inoculation to C57 / BL mice continuous intraperitoneal injection of drug 7d to explore the survival of tumor-bearing mice Eb, Tumor growth rate, infiltration level, cell morphology, cell cycle and apoptosis. Results: Eb could significantly inhibit tumor growth and invasion (high-dose Eb tumor inhibition rate was 80.31%), prolong the survival of tumor-bearing mice; morphological observation found that after the administration of Eb tumor cells concentrated deep staining, mitotic cells decreased; Flow cytometry results show that, Eb can promote tumor cell apoptosis. CONCLUSION: The new organic selenium compound Eb can significantly inhibit the growth and invasion of LLC in C57 / BL mice, promote the apoptosis of tumor cells and have strong anti-tumor activity.
