目的:观察玉液汤加减对高脂饲养大鼠血浆肿瘤坏死因子α(TNF-α)、纤溶酶原激活物抑制剂-1(PAI-1)浓度的影响。方法:♂SD大鼠75只,随机分为正常对照组(第1组)、单纯高脂组(第2组)和玉液汤加减低、中、高剂量干预组(第3~5组)。6个月后测定空腹血糖、血浆胰岛素水平及生化指标,计算胰岛素抵抗指数(HOMA-IR),胰岛素敏感指数(ISI),观察玉液汤加减对大鼠血浆TNF-α、PAI-1浓度及胰岛素抵抗的影响。结果:单纯高脂组的血浆TNF-α、PAI-1浓度水平均较正常对照组明显升高(P<0.01),并出现胰岛素抵抗;玉液汤加减中剂量组、高剂量组的血浆TNF-α、PAI-1浓度较单纯高脂组均明显降低(P<0.01),胰岛素敏感性明显提高。结论:大鼠高脂饮食可引起血浆TNF-α和PAI-1浓度增高,同时出现胰岛素抵抗;玉液汤加减在降低血浆TNF-α和PAI-1浓度的同时也改善了高脂饮食大鼠胰岛素抵抗。因此玉液汤加减对防治高脂饮食胰岛素抵抗及临床治疗2型糖尿病具有巨大的开发应用价值。
Objective: To observe the effects of Yuye Decoction on plasma tumor necrosis factor-α (TNF-α) and plasminogen activator inhibitor-1 (PAI-1) concentrations in rats fed with high-fat diet. Methods: Seventy-five Sprague-Dawley rats were randomly divided into normal control group (Group 1), simple high-fat group (Group 2) and Yuye Decoction plus low-, medium-, and high-dose intervention groups (Groups 3 to 5). After 6 months, the fasting blood glucose, plasma insulin and biochemical indexes were measured. The HOMA-IR and ISI were calculated. The concentrations of TNF-α and PAI-1 in plasma of rats were observed after the addition and subtraction of Yuye Decoction. The effect of insulin resistance. Results: Plasma TNF-α and PAI-1 levels in the high-fat group were significantly higher than those in the normal control group (P<0.01), and insulin resistance appeared. Plasma TNF in the middle-dose and high-dose groups of Yuye Decoction was increased. The concentrations of α-PAI-1 and PAI-1 were significantly lower than those in the high-fat group (P<0.01), and insulin sensitivity was significantly increased. Conclusion: Rats with high-fat diet can increase plasma concentrations of TNF-α and PAI-1, as well as insulin resistance. Yuyin decoction can reduce the concentration of plasma TNF-α and PAI-1 and improve the high-fat diet in rats. Insulin resistance. Therefore, the addition and subtraction of Yuye Decoction is of great value in the development and application of high-fat diet insulin resistance and clinical treatment of type 2 diabetes.
