目的探讨肝细胞肝癌(HCC)中端粒酶逆转录酶(hTERT)与c-myc蛋白表达情况及其相关性。方法采用免疫组化SP法检测52例HCC癌组织及相应癌旁组织中hTERT与c-myc蛋白表达,并用TRAP-ELISA法检测上述组织中的端粒酶活性。结果HCC组织中hTERT与c-myc蛋白阳性率分别为86.5%(45/52)和78.8%(41/52),它们与相应癌旁组织比较差异有统计学意义(P<0.01);HCC癌组织中端粒酶活性阳性率为80.8%(42/52),明显高于相应癌旁组织(15.4%,8/52);HCC癌组织中端粒酶活性、hTERT及c-myc蛋白阳性表达率两两均呈显著正相关(r=0.761,P<0.001;r=0.654,P<0.001;r=0.486,P<0.001)。结论HCC癌组织中hTERT与c-myc蛋白过度表达,两者相互作用而共同参与肝癌端粒酶活性的调控。
Objective To investigate the expression of telomerase reverse transcriptase (hTERT) and c-myc in hepatocellular carcinoma (HCC) and its correlation. Methods The expressions of hTERT and c-myc in 52 HCC tissues and corresponding paracancerous tissues were detected by immunohistochemical SP method. The telomerase activity in these tissues was detected by TRAP-ELISA. Results The positive rates of hTERT and c-myc protein in HCC tissues were 86.5% (45/52) and 78.8% (41/52), respectively, which were significantly different from those in corresponding paracancerous tissues (P <0.01) The positive rate of telomerase activity in the tissue was 80.8% (42/52), which was significantly higher than that in the corresponding paracancerous tissue (15.4%, 8/52). The positive expression of telomerase activity, hTERT and c-myc in HCC tissues There was a significant positive correlation between the two rates (r = 0.761, P <0.001; r = 0.654, P <0.001; r = 0.486, P <0.001). Conclusions The expression of hTERT and c-myc in HCC tissues is overexpressed, and the two interact with each other to participate in the regulation of telomerase activity in HCC.
