反相高效液相色谱法测定人体血浆中游离羟脯氨酸

羟脯氨酸是人体胶原蛋白的特有氨基酸。在胶原蛋白代谢异常时,必然反映在血羟脯氨酸的变化上,所以测定病人在治疗过程中血羟脯氨酸含量变化,对中西医结合抗肝脏纤维化治疗过程中的疗效考核,是具有一定临床意义的。我们根据目前国内供货来源,在Peter等所用自动荧光氨基酸分析法基础上,改用反相高效液相色谱法,进行了探索,取得了比较满意的结果。 层析条件 C_(18)H_(37)键合相层析柱:150×4.6mmI.D.;C_(18)H_(37)键合相预柱:50×4.6mmI.D.;荧光检测器:λ_(ex)为347nm,λ_(em)为455nm:柱温:25℃:流动相:0.03M柠檬酸三钠缓冲液(pH=7): Hydroxyproline is a unique amino acid of human collagen.
