目的观察血红素加氧酶-1(HO-1)重组乳酸乳球菌同一剂量不同灌胃次数对正常大鼠肠黏膜屏障的影响。方法将40只健康雄性SD大鼠随机分为:HO-1重组乳酸乳球菌灌胃一次组(HO1t组,n=8)、HO-1重组乳酸乳球菌灌胃二次组(HO2t组,n=8)、HO-1重组乳酸乳球菌灌胃三次组(HO3t组,n=8)、PBS溶液灌胃一次组(PBS1t组,n=8)和乳酸乳球菌灌胃一次组(LL1t组,n=8)。各组灌胃剂量的容积均为1ml,每次灌胃间隔24h,在末次灌胃24h后取回肠组织,检测肠组织Chiu’s 6级评分、髓过氧化物酶(MPO)活性,同时从右心室抽取血液行细菌学检查。结果各组肠黏膜Chiu’s评分比较差异无统计学意义;与LL1t组比较,HO2t组和PBS1t组的肠组织内MPO活性较高,但比较没有统计学意义;HO2t组有1例、HO3t组有2例血培养阳性,其余各组血培养阴性。结论 HO-1重组乳酸乳球菌对正常大鼠灌胃,不会随着灌胃次数增加而影响肠黏膜屏障的功能。
Objective To observe the effect of the same dose of heme oxygenase-1 (HO-1) recombinant Lactococcus lactis on the intestinal mucosal barrier in normal rats. Methods Forty healthy male Sprague-Dawley rats were randomly divided into two groups: HO-1 recombinant Lactococcus lactis (HO1t group, n = 8), HO-1 recombinant Lactococcus lactis (N = 8, HO-1) and Lactococcus lactis (LL1t group, n = 8) n = 8). The volume of intragastric dose in each group was 1ml. After intragastric administration for 24 hours, the intestinal tissues were removed 24 hours after the last intragastric administration, and the Chiu’s 6th grade and MPO activity were determined. At the same time, Ventricular blood was taken for bacteriological examination. Results There was no significant difference in the Chiu’s score of intestinal mucosa between groups (P> 0.05). Compared with LL1t group, MPO activity in intestinal mucosa of HO2t group and PBS1t group was higher, but there was no statistical significance; 1 case in HO2t group and 2 cases in HO3t group Blood culture positive, the rest of the blood culture negative. Conclusion HO-1 recombinant Lactococcus lactis can be administered to normal rats without affecting the function of intestinal mucosal barrier with the increase of gavage times.
