目的:观察补肾活血方补肾活血方对前列腺增生大鼠膀胱逼尿肌缝隙连接蛋白43(Cx43)mRNA及蛋白表达的影响。材料与方法:清洁级雄性Wistar大鼠48只,随机分为正常组、模型组、模型加补肾活血方组、模型加坦索罗辛组四组。除正常组外,其它实验大鼠以去势加睾酮注射法复制大鼠BPH模型。于去势第一天起,各组大鼠分别给予相应药物灌胃。模型+补肾活血方按1mL/100g/日体重予补肾活血方(由淫羊藿、枸杞子、穿山甲、丹参、菟丝子等组成,含生药量2.34g/ml)灌胃,模型加坦索罗辛组按1.8μg/100g/日体重予坦索罗辛灌胃。其余各组予等体积生理盐水灌胃。均每日1次,连续38天后检测各组大鼠膀胱逼尿肌组织Cx43 mRNA及蛋白表达。结果:与空白组相比,模型组Cx43 mRNA表达明显上升(2.08 vs 30.69,P<0.01),坦索罗辛及补肾活血方组Cx43 mRNA表达与空白组相比也有升高,但无统计学差异(16.05,12.74,2.08,P>0.05)。与模型组相比,补肾活血方(12.74 vs 30.69,P<0.01)Cx43 mRNA表达明显下降。与空白组相比,模型组及补肾活血方、坦索罗辛等各组Cx43蛋白表达明显上升(分别为1.44,3.89,1.86,2.73),而补肾活血方、坦索罗辛各组与模型组相比,Cx43蛋白表达均明显下降(P<0.01)。结论:补肾活血方组可以明显降低BPH模型大鼠膀胱组织Cx43 mRNA及蛋白表达,这可能是补肾活血方有效改善BPH继发膀胱改变的机制之一。
Objective: To observe the effect of Bu Shen Huo Xue Fang Bu Shen Huo Xue Fang on mRNA and protein expression of connexin 43 (Cx43) in detrusor of benign prostatic hyperplasia rats. MATERIALS AND METHODS: Forty-eight male Wistar rats were randomly divided into four groups: normal group, model group, model plus Bushen Huoxue recipe group and model plus tamsulosin group. In addition to the normal group, other experimental rats with castrate plus testosterone injection of rat BPH model. On the first day of castration, the rats in each group were given the corresponding drugs respectively. Model + Bushen Huoxue Fang was given according to 1mL / 100g / day body weight to Bushen Huoxue prescription (composed of Epimedium, wolfberry fruit, pangolin, Salvia miltiorrhiza, dodder and other ingredients containing crude drug amount 2.34g / ml) Group according to 1.8μg / 100g / day weight to tamsulosin intragastric administration. The remaining groups to the same volume of normal saline gavage. Each group was given once a day for 38 days continuously to detect the expression of Cx43 mRNA and protein in detrusor tissue of rats in each group. Results: Compared with the blank group, the expression of Cx43 mRNA in the model group was significantly increased (2.08 vs 30.69, P <0.01), while the expression of Cx43 mRNA in tamsulosin and Bushen Huoxue prescription group was also increased compared with the blank group (16.05,12.74,2.08, P> 0.05). Compared with the model group, the expression of Cx43 mRNA of Bushen Huoxue prescription (12.74 vs 30.69, P <0.01) was obviously decreased. Compared with the blank group, the expressions of Cx43 protein in the model group, Bushen Huoxue Recipe and tamsulosin group were significantly increased (1.44, 3.89, 1.86, 2.73, respectively) Cx43 protein expression was significantly decreased (P <0.01). Conclusion: Bushen Huoxue Recipe can significantly reduce the expression of Cx43 mRNA and protein in bladder tissue of BPH model rats, which may be one of the mechanisms that Bushen Huoxue Decoction can effectively improve secondary bladder changes of BPH.
