博莱霉素组份A_2(55~70%)和B_2(25%)用一般方法难以分离。反相高效液相层析法可成功分离博莱霉素,但需要4~5小时。作者利用博莱霉素的强阳离子性质,采用反相离子对高压液相层析法,可快速地分离博莱霉素组份。高压液相色谱仪为ALC202/6000型。备有440型吸收检测器及450型可变波长吸收检测器,于紫外280~29O毫微米处测量。μBondapak C_(18)反相柱(300×4.6毫米内径),于室温操作。移动相为5毫克分子正庚烷磺酸的50%甲醇液,用氢氧化铵调节至pH8.3,流速0.5~1.5毫升/分。
Bleomycin components A 2 (55-70%) and B 2 (25%) are difficult to separate by conventional methods. Bleomycin was successfully separated by RP-HPLC, but it took 4 to 5 hours. Using the strong cation nature of bleomycin, the authors can rapidly separate the bleomycin fraction using reversed-phase ion-pair high pressure liquid chromatography. High pressure liquid chromatography for the ALC202 / 6000 type.
