为了解肾综合征出血热乳鼠脑Ⅱ型纯化疫苗侯选病毒株R2 2 的分子生物学特征 ,应用PCR方法克隆了R2 2株的全S基因 ,并测序分析比较。结果R2 2 株的S片段的全基因序列共 1774个核苷酸 ,编码 42 9个氨基酸。与汉坦病毒I型 76 118株核苷酸和氨基酸同源率分为 73 7%、83 3 % ,而与同型的SR 11株则高达 95 6 %、97 9%。核苷酸序列比较发现R2 2 在 170 1~ 170 9有一ACCTCCTA 7个碱基的插入 ,175 6~ 176 0处有一 5个碱基的缺失。应用DNASTAR软件 ,绘出了R2 2 株病毒S基因编码的蛋白质的二级结构。
In order to understand the molecular biological characteristics of candidate glycoprotein R2 2 of neonatal rat model of hemorrhagic fever with neonatal hemorrhagic fever, the whole S gene of R2 2 strain was cloned by PCR and sequenced and compared. Results The whole gene sequence of the S segment of R2 2 strain was 1774 nucleotides in length, encoding 429 amino acids. The nucleotide and amino acid identities of 76 118 strains of Hantaan virus were 73 7% and 83 3%, respectively, while they were 95 6% and 97 9% higher than those of the same type SR 11 strains. The comparison of nucleotide sequences showed that R2 2 had a 7 base insertion of ACCTCCTA at 170 1 ~ 170 9 and a 5 base deletion at 175 6 ~ 176 0. The DNASTAR software was used to map the secondary structure of the protein encoded by the R2 gene of the virus S gene.
