针对市售枸杞提取物中多糖含量难以确定的问题,建立了先用淀粉葡萄糖苷酶脱除麦芽糊精,再用高效凝胶色谱分离定量的检测方法,经分析验证淀粉葡萄糖苷酶降解麦芽糊精的同时对枸杞多糖含量并无显著显影响,高效凝胶色谱可以实现不同分子量范围多糖的精确定量。检测结果表明,枸杞提取物中多糖含量为12%,相对分子质量范围为10 000~60 000 U,低聚糖含量较高为51.3%。
Aiming at the problem that the content of polysaccharide in the commercially available Chinese wolfberry extract is difficult to be determined, a method for separating maltodextrin by using starch glycosidase and establishing a quantitative detection method by high-performance gel permeation chromatography was established. At the same time, the content of Lycium barbarum polysaccharides was not significantly affected by high-performance gel chromatography, and the precise quantification of polysaccharides with different molecular weight range was achieved by high-performance gel chromatography. The results showed that the polysaccharide content of Lycium barbarum extract was 12%, the relative molecular mass range was 10 000 ~ 60 000 U and the content of oligosaccharides was 51.3%.
