目的:综述分析国内外增强自然杀伤细胞(naturalkillercell,NK)对肿瘤杀伤活性的研究现状,为基础与临床研究提供研究方向、思路和资料。方法:应用PubMed和万方数据库等检索系统,以NK细胞、肿瘤和细胞毒等为关键词,检索1998-01-2008-07与NK细胞肿瘤杀伤相关的文献。纳入标准:NK细胞在肿瘤免疫治疗中的基础与临床研究。粗选230篇文献,最后34篇纳入综述分析。结果:NK细胞是一种独特的淋巴细胞,对靶细胞的识别无MHC限制性,无需预先致敏即可直接杀伤肿瘤细胞,也可分泌细胞因子调节其他免疫细胞的功能,是机体天然免疫的主要承担者,也是获得性免疫的核心调节细胞,故NK细胞在抗肿瘤免疫中的地位越来越受到重视,随之增强NK细胞肿瘤杀伤活性的研究也逐步深入。可以通过细胞因子、单抗及多功能抗体介导NK细胞活化,通过调节抑制性信号和活化性信号传递加强NK细胞活性,通过免疫药物刺激及基因修饰提高NK细胞功能等多种途径增强NK细胞的肿瘤杀伤作用。结论:研究增强NK细胞杀伤活性的方法,将促进NK细胞在肿瘤免疫治疗中的应用。
OBJECTIVE: To summarize and analyze the current situation of natural killer cells (NK) on tumor killing activity at home and abroad, and to provide research directions, ideas and data for basic and clinical research. Methods: Using search systems such as PubMed and Wanfang database, the articles related to NK cell tumor killing were retrieved from January 1998 to July 2008 with the keywords of NK cells, tumor and cytotoxicity. Inclusion criteria: Basic and clinical studies of NK cells in tumor immunotherapy. Crude selection of 230 articles, the last 34 included in the review. Results: NK cells were a kind of unique lymphocytes, which had no MHC restriction on the recognition of target cells, could directly kill tumor cells without pre-sensitization, and could also secrete cytokines to regulate the function of other immune cells. The main commitment, but also the core of adaptive immunity regulatory cells, so the status of NK cells in anti-tumor immunity more and more attention, followed by enhanced NK cell tumor killing activity of the research has gradually deepened. NK cells can be activated through cytokines, monoclonal antibodies and polyfunctional antibodies, NK cells can be enhanced through regulating inhibitory and activating signal transduction, and NK cells can be enhanced by immune drug stimulation and gene modification to improve NK cell function Tumor killing effect. Conclusion: To study the method of enhancing NK cell killing activity will promote the application of NK cells in tumor immunotherapy.
