目的:用不同培养基、不同浓度乙醇沉淀分离得到的蝉花多糖的浓度和清除自由基DPPH的能力不同。方法:将不同液体培养基体外发酵培养的蝉花发酵液,经分级醇沉,离心取粗多糖,按照苯酚硫酸法测出蝉花发酵液胞外总糖含量,其抗氧化活性用清除1,1-二苯基苦基苯肼(DPPH)有机自由基和羟自由基(OH)能力的方法进行评价。结果:不同培养基发酵培养的经不同乙醇量沉淀提取的蝉花胞外多糖的总糖含量不同,其相应清除DPPH有机自由基和羟自由基(OH)的能力也有较为明显的不同。其中方法2培养基用75%乙醇提取的多糖含量最高为96.43%,其清除DPPH有机自由基的能力的也最高为63.68%。结论:蝉花多糖有一定清除有机自由基能力,方法2培养基经75%乙醇提取到的蝉花多糖的含量最高,清除DPPH有机自由基的能力也最高。
OBJECTIVE: The concentrations of cicadaeana polysaccharide isolated from different media and different concentrations of ethanol were different from those of free radical DPPH. Methods: The culture broth of cicadas cv. Phylloxera in different liquid culture medium was graded by alcohol precipitation and crude polysaccharide was centrifuged. The total extracellular sugar content of cicadae fermentate liquid was measured by phenol sulfuric acid method. 1-diphenyl picrylhydrazide (DPPH) organic free radicals and hydroxyl radical (OH) capacity of the method of evaluation. Results: The total sugar content of cicadae extracellular polysaccharides extracted by different ethanol precipitates in different culture medium was different, and their abilities to scavenge DPPH organic radical and hydroxyl radical (OH) also had obvious differences. Among them, the content of polysaccharides extracted by 75% ethanol was 96.43% in the medium of method 2, and the highest ability of clearing DPPH organic free radicals was also 63.68%. CONCLUSION: Cicadae polysaccharide has the ability to scavenge organic free radicals. The content of Cicadaema polysaccharides extracted by 75% ethanol in method 2 medium is the highest, and the scavenging capacity of DPPH organic free radicals is the highest.
