目的探讨适宜浓度α-亚麻酸(ALA)对卵胞浆内单精子注射(ICSI)周期中人未成熟卵母细胞体外成熟、受精和胚胎发育的影响。方法选取2014年10月~2015年2月在该院接受ICSI治疗的不孕患者150名为研究对象,将其ICSI周期中未成熟卵母细胞随机分为ALA组(A组)、成熟卵泡液组(B组)和空白对照组(C组)。3组细胞在人未成熟卵母细胞体外成熟培养(IVM)培养液中培养24 h,观察未成熟卵母细胞体外成熟情况及胚胎发育情况。同时测定卵母细胞培养基中超氧化物歧化酶(SOD)总活力和丙二醛(MDA)含量。结果 A组24 h成熟率高于B组,B组高于C组,差异均有统计学意义(P<0.05);A组和B组卵母细胞ICSI后2PN卵裂率高于C组,差异有统计学意义(P<0.05);3组2PN受精率、可利用胚胎率、囊胚形成率和SOD活力两两比较,差异无统计学意义(P>0.05);A组MDA含量低于B组和C组,差异均有统计学意义(P<0.05)。结论 ALA通过降低氧化应激水平促进卵母细胞体外成熟,提高胚胎发育潜能。
Objective To investigate the effect of appropriate concentration of α-linolenic acid (ALA) on in vitro maturation, fertilization and embryo development of human immature oocytes in the intracytoplasmic sperm injection (ICSI) cycle. Methods A total of 150 infertile patients receiving ICSI from October 2014 to February 2015 in our hospital were enrolled in this study. Immature oocytes in the ICSI cycle were randomly divided into ALA group (A group), mature follicular fluid Group (group B) and blank control group (group C). Three groups of cells were cultured in IVM medium of immature oocytes for 24 hours to observe the in vitro maturation and embryo development of immature oocytes. The total superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in oocyte medium were also determined. Results The maturation rate of 24 h in group A was higher than that in group B, while that in group B was higher than that in group C (P <0.05). The 2PN cleavage rate of ICSI in group A and group B was higher than that in group C, The difference was statistically significant (P <0.05); 3P 2PN fertilization rate, available embryo rate, blastocyst formation rate and SOD activity of any pairwise comparison, the difference was not statistically significant (P> 0.05) B and C groups, the differences were statistically significant (P <0.05). Conclusion ALA can promote oocyte maturation in vitro and increase embryonic development potential by reducing oxidative stress.
