妊娠早期子宫内产生的前列腺素可能是胎体生存的必需物质。本实验用离体培养法测定牛子宫内膜是否会改变胚泡对〔~3H〕-花生四烯酸(〔~3H〕-AA)的代谢。采用人工授精妊娠19天牛的子宫,取出其胚泡,并将其子宫内膜切成近18mm~3大小,分成四组体外培养。第一组单独培养胚泡;第二组单独培养子宫内膜;第三组胚泡和子宫内膜共同培养;第四组胚泡的培养液一半取自子宫内膜培养液。还有一组培养液做对照。所有的都放在15ml改良的最小量基本培养液中,通以50%N_2∶45%O_2∶5%CO_2,在37℃培养24小时。每一组都加入5μCi的〔~3H〕-AA和200μg无放射活性的AA。培养结束在4℃离心分离开组织和培养液,保存于-20℃中。
Prostaglandins produced in the uterus during early pregnancy may be essential for the survival of the carcass. In this study, in vitro culture was used to determine whether the bovine endometrium could alter the metabolism of [~ 3H] -arachidonic acid ([~ 3H] -AA) in blastocysts. Artificial insemination pregnancy 19 days cow uterus, remove the blastocyst, and its endometrium cut into nearly 18mm ~ 3 size, divided into four groups in vitro. The first group was cultured in blastocyst alone; the second group was cultured in endometrium alone; the third group was co-cultured with blastocyst and endometrium; and the fourth group of blastocyst culture medium was taken from endometrial culture medium. There is also a group of culture medium control. All were placed in 15 ml modified minimal minimal medium and incubated for 24 hours at 37 ° C with 50% N_2: 45% O_2: 5% CO_2. 5μCi of [~ 3H] -AA and 200μg of non-radioactive AA were added to each group. After the end of the culture, the tissue and the culture medium were centrifuged at 4 ° C and stored at -20 ° C.
